When you go to the site in IIS Manager do you see the rewrite rules (specifically the api one) in the URL Rewrite module? If so then check the various logs (general server logs, webserver error logs, PHP error logs, MySQL/MariaDB error logs, osTicket System Logs, Browser Console logs, etc.) for any related errors.
