I have added 4 form variables to tickets. I am using those variables for canned messages. BUT is there a way, to make an agent fill the custom variable BEFORE he uses a canned message, that uses this variable? Something like a requester pops up, when a canned message is used and the variable is not set.

    No, if not set it will simply show the variable untouched or replaced with an empty string.
