I also tried user separate authenticator and giving 2FA password that also didn't help requesting guidance

Sounds like a firewall issue or something. Make sure you can telnet to that host and port from your webserver. If you can then you will need to do some additional debugging. From osTicket side of things we just cannot connect to the given hostname/port.
